Can I calculate an area containing a hole or holes?

No, but with drawing layers it’s as simple as subtracting the area of the hole or holes, from the area of it’s surrounding shape.

If you had a complex area calculation containing multiple holes, say an image of swiss cheese, and you needed the area of the cheese. Draw each hole on its own layer and sum the area of all those layers together by selecting the eye symbol in the results bar, found in the bottom right corner.

Take a note of the summed area of the holes then draw around the perimeter of the cheese itself. Simply subtract the combined area of the holes from the area of the cheese.
